
Hello friends, welcome to the channel solution Today we will show the the procedure for shinoda test Which is used for the detection of flavonoids in any plant materials Here we have taken orange peal powder For the detection of flavonoids First of all transfer small amount of orange peal powder into the conical flask and then add ethanol 95% for the preparation of ethanolic extract after the addition of ethanol into the conical flask shake the conical flask so that ethanol may mix completely with the all organge peal powder after shanking cover the conical flask with the cork and then leave the content of the flask for 30 mints After 30 mint filter the content of conical flask through the normal filter paper and then collect the filtrate into the beaker Here is the ethanolic extract we have collected after the filtration in which we have to perform the Shinoda test for flavonoids take about 2-3 ml of ethanolic extract of orange peal which we have prepared and then transfer into the test tube after the addition of ethanolic extract into the test tube Then add few drops of concentrated hydrochloric acids Now add 2 pieces of magnesium ribbon After the addition of these two if the colour changes to orange red, pink or purple colour Then it indicates the presence of flavonoids Here the colour changes to red, it means in our extract flavonoids is present Means Shinoda test is positive The principle of Shinoda test is that Flavonoid which is present in the extract after the addition of magnesium ribbon and HCl It reduces to anthocyanidins And due to formation of this anthrocynidine Colour of the extract changes to red Now we are going to perform another test for flavonoids That is alkaline reagent test For this test take about same ethanolic extract of orange peel powder which we have prepared earlier And with the help of dropper transfer it into a test tube Transfer the same amount of ethanolic extract into another test tube And this test tube is taken for the reference purpose After the addition of extract into the test tube Add drop by drop 2% of NaOH solution in both the test tube After the addition of NaOH Solution The colour of extract changes to intense yellow colour After the formation of yellow color after the addition of NaOH solution in both the test tube then we have to add dilute Hcl with the help of dropper transfer few drops of dilute Hcl in the left test tube we can see the colour Which was formed after the addition of NaOH Solution Starts disappear after the addition of dilute Hcl And will completely disappear after few seconds We can see that colour completely disappears This confirms the presence of flavonoid in our extract We can compare the result with the extract In the test tube of the right side Thank you for watching this video.
